Article history: Received on: 05/11/2012 Revised on: 29/11/2012 Accepted on: 18/12/2012 Available online: 28/02/2013 Anisodus acutangulus hairy roots were grown in N6 medium which was optimal for growth and alkaloid production. The cell biomass and alkaloid yield reached 2.7 g l and 3.9 mg l (dry weight) respectively in sucrose medium, higher than those obtained in other carbon source media. 90 mM nitrogen (NH4/NO3 = 4:1) gave the highest cell yield (4.5 g l) and the maximum alkaloid production (9.9 mg l). The cell yield (4.1 g l) of hairy roots grown at pH 6.5 was 2 times higher than that at pH 4.5. However, the maximum alkaloid production (7.2 mg l) was yielded at pH 4.5.

Agrobacterium rhizogenes (pRi), a causative agent of hairy root disease, effectively induces hairy root formation in a variety of plant species. In our study four bacterial strains AR15834, A4, 9435 and C318 and three explants types leaf, stems and roots, were examined. Hairy roots were induced from roots, stems and leaf explants. The highest transformation efficiency of 77% was achieved by usi...

BACKGROUND Tropane alkaloids (TA) including anisodamine, anisodine, hyoscyamine and scopolamine are a group of important anticholinergic drugs with rapidly increasing market demand, so it is significant to improve TA production by biotechnological approaches. Putrescine N-methyltransferase (PMT) was considered as the first rate-limiting upstream enzyme while tropinone reductase I (TRI) was an i...

Plants are a potential source of a large number of valuable secondary metabolites. In vitro cultures are being considered as an alternative to agricultural processes for studying valuable secondary metabolites. In this way, nutritive factors are important parameters influencing the production of these compounds in plants. Effects of nitrate concentrations (KNO3) on the production of two tropane...

Background & Aim:A new sustainable tissue-based system is presented by plant hairy roots, preserving all of the several specialized types of cell with critical roles in allowing bioactive secondary molecules to be synthesized more consistently as usual. The system is also essential for studying the production of alkaloid in culture. Experimental: The ...

The (-)-hyoscyamine, atropine and scopolamine (hyoscine) are three valuable tropane alkaloids while scopolamine is the most important member of this group for the pharmaceutical industry due to its higher demand compared to hyoscyamine and atropine. Scopolamine is an anticholinergic reagent with several therapeutic applications. In the current study, the hairy roots culture of Daturametel was u...
